This research has demonstrated the partial purification of hyaluronic acid from human umbilical cord. This is done by a series of steps, making use of enzymatic digestion of the umbilical cord, repeated alcohol salt precipitation of the enzyme extracts, and gel filtration and ion exchange steps. After the ion exchange step, the hyaluronic acid elutes as several sharp peaks, reflecting a series of hyaluronate species of different molecular weights.
The hyaluronic acid we have produced still contains some amount of proteins. Further work is required to achieve a preparation of greater purity for use in opthalmoic applications. The hyaluronic acid must be characterized as to its velocity, transparency and miscibility. Spectral analysis using infrared spectroscopy should also be attempted. Characterization by circular dichroism or optical rotatory dispersion, if available, could also be done. (Author)
The study aims to: 1) produce ophthalmic grade hylauronic acid (HA) from human umbilical cord (HUC); 2) characterize the preparation of HA according to standard criteria; and 3) evaluate the effectiveness of the preparation in viscosurgery, initially through animal experiments and then in human trials.
